Improving your CTR - untick Online Advertiser Sign-Up
I requested some assistance on DP earlier this week in regards to the placement of adsense to improve CTR. An interesting tip that I hadn't thought of related to the text that appears in the bottom right hand corner of the Google Ads, namely Advertise on this site.
The purpose of that text link is that it allows advertisers to advertise specifically on your site, by bidding on keywords available that are suitable for the sites content. The catch here is that you make nothing from the advertiser clicking that, even though you are generating revenue for Google by having people sign up for their adsense program (maybe not in all cases as some who click would be existing users, but nonetheless...)
The problem with that text link is that it leaves the viewer in no doubt that they are looking at advertisements, which could decrease your potential CTR. I was advised that removing this text, would take this variable out of the equation and as such the CTR may increase. I've decided to give it a go and see what results I get.
If you want to remove the Advertise on this site text link, follow these steps:
1 Log in to your AdSense account.
2 Under the My Account tab, visit the Account Settings page.
3 Scroll down to the Onsite Advertiser Sign-up section (right at the bottom) and click 'edit.'
4 Untick the Include me in Onsite Advertiser Sign-Up check box next to Subscription:
5 Click Save changes.
It may take some time for the change to take effect on your site so be patient.

It did take some time for that link to disappear (about 48 hours).
Basically what happens is the Ads By Google link shifts over to the right hand side to replace it.
When the Advertise on this site link is in place the Ads By Google link sites on the left hand side.
So having only 1 link gives might give better click through opportunity with those left hand ads that aren't surrounded by the google ad text - again we'll wait and see
On the flip side. That Advertise on this site link, when removed, stops people that want to advertise on your site from doing so directly. Personally I prefer this removed as the sites that were listing on here some of the time were totally unrelated, and I don't like their unclickable ads sitting here absorbing the adspace, I'd rather have randoms.
I also think this could be good for preventing possible competition from advertising directly on your site. I know there are competitive filters in the Adsense configuration, but that only helps you remove competitors that you are aware of. What happens to those that spring up that you are unaware of? They have a nice fat link saying "advertise on this site" and they cut you out by going straight to google.

The last 2 days my CTR has been up about 0.3% on what it normally is. Marginal I know, but depending on what your earning that can be quite a bit.

I have to agree, and was going to mention the fact that the 'ads by gooooogle' link moves as being a major factor. People read top to bottom, left to right in the Western world, so that black text ads by google was eyecatching. Moved over to the right it is unobtrusive.

It's on the left with rectangles.
Don't forget to disallow the Google sites from showing up in the AdSense ads. If people click them and register for AdSense or AdWords, you get a percentage of one click. It's better to put AdSense and AdWords buttons on the site and get a lot more money for those registrations.
I've just redone the positioning of the AdSense stuff on my webworkshop.net site, and my income immediately increased by between 50% and 90%, so positioning does help a lot.
